Toxicity to blood cells of SD rats from sub-chronic exposure to DEHP

Abstract: OBJECTIVE: To observe toxicological effects in blood cells in rats after treatment with different doses (0,5,50,500,2 500 mg/kg) DEHP. METHODS: Rats were divided into control and four treatment groups. The treatment groups were exposed to four concentrations of DEHP (5,50,500 and 2 500 mg/kg) for 90 days. At the end of the exposure,whole blood samples were taken to measure the changes of various types of blood cells. RESULTS: Compared with the control group,the leukocyte parameters for the 2 500 mg/kg dose groups were:eosinophils decreased significantly (P < 0.01),the total number of white rats significantly decreased (P < 0.05). Lymphocytes in 500 and 2 500 mg/kg groups were significantly increased (P < 0.05). In the erythrocyte parameters,the total number of red blood cells,hemoglobin (HGB) and hematocrit (HCT) in the 2 500 mg/kg group were significantly decreased (P < 0.01) while the levels of hemoglobin (HGB) in 500 and 2 500 mg (P < 0.05 or P < 0.05). Mean hemoglobin content (MCH) in 500 and 2 500 mg/kg groups decreased significantly (P < 0.05 or P < 0.01). The measured hemoglobin concentration (CHCM) decreased significantly (P < 0.05 or P < 0.01). On the other hand,DEHP had little effect on platelet parameters. CONCLUSION: DEHP exposure affected blood cells of rats under certain dosages.
