氯化镉对小鼠睾丸指数及其线粒体ATPase6、D-Loop基因突变的影响

Effects of Cadmium Chloride on Index and mtATPase6 and D-Loop Mutations of Mice Testicle

摘要: 背景与目的: 研究氯化镉对睾丸指数及睾丸细胞线粒体ATPase6、D-Loop基因突变的影响。 材料与方法: 取40只体重(19.5±2.5)g的雄性ICR小鼠,随机分为4组,每组10只,分别以1、5、10 μmol/kg氯化镉腹腔注射,隔天注射1次,共10次,同时设阴性对照组(生理盐水)。第21天取小鼠双侧睾丸称重并计算睾丸指数;提取睾丸组织基因组DNA,PCR扩增线粒体ATPase6、D-Loop基因,纯化后测序分析基因突变。 结果: 10 μmol/kg氯化镉组睾丸指数显著低于对照组(P<0.01),未检测到小鼠睾丸细胞线粒体ATPase6、D-Loop基因的突变。 结论: 10 μmol/kg浓度的氯化镉可致小鼠睾丸指数降低,氯化镉短期处理(21 d 内)未能引起小鼠睾丸线粒体ATPase6、D-Loop基因突变。睾丸指数与细胞线粒体ATPase6、D-Loop基因突变可能不存在相关性。

Abstract: BACKGROUND AND AIM: To study testis index and mtATPase6 and D-Loop point mutations of mice testicle after exposure to cadmium. MATERIALS AND METHODS: 40 male mice were divided into 4 groups with 10 animals per group (weight:19.5±2.5 g).3 groups were treated with intraperitoneal cadmium chloride of 1,5,10 μmol/kg,once every 2 days,10 times in total,and one normal saline control group. After exposure to cadmium chloride for 20 days, we determined the testis index. The mtATPase6 and D-Loop genes were amplified by PCR from the genomic DNA of mice testicle tissue, with their mutant sites sequenced. RESULTS: The testis index of mice in the 10 μmol/kg cadmium chloride group were lower than that of the control group(P<0.01). No mtATPase6 or D-Loop mutation was found in the murine spermatogenic cells. CONCLUSION: Mice testis index was lower by 10 μmol/kg cadmium chloride. But cadmium chloride couldn't induce mtATPase6 and D-Loop mutations in the mice testicle. Testis index and mtATPase6 and D-Loop point mutations did not seem to be related.
